You should make a backup of your site at least once a week if it is a busy site, also it is very important that you make backups of both the database and files before making any structure change like a version upgrade or before adding new modules, that way if something goes wrong you can restore your site to how it was before, before clicking on Backup DB you should click on Optimize DB to clean out any useless data that may be taking up space.
